Description: Developmental diseases are a set of heterogeneous disorders characterized by broad clinical developmental features, comprising sensorineural hearing loss (SNHL). Chromatinopathies are part of these developmental diseases caused by gene mutations encoding proteins with roles in chromatin conformation and/or function. Cornelia de Lange syndrome (CdLS) is one of these Chromatinopathies. About 65% of diagnosed cases result from mutations in cohesin loading factor Nipped-B-like (NIPBL). To better understand SNHL in NIPBL-mutated CdLS patients, we aim to use patient-derived induced pluripotent stem cells (iPSC) -differentiated in otic progenitors and otic cells- as a monolayer or 3D organoids and a Nipbl heterozygous (Nipbl+/-) mouse model.
